چکیده

SpiC is a virulence factor encoded within Salmonella pathogenicity island 2 (SPI-2). We have previously reported that infection of macrophages with Salmonella enterica serovar Typhimurium results in the SPI-2-dependent activation of the mitogen-activated protein kinase (MAPK) signalling pathways, leading to the expression of suppressor of cytokine signalling (SOCS)-3, which is involved in the inhibition of cytokine signalling. Here, we investigated the mechanism by which SpiC mediates the activation of signal transduction pathways in macrophages. Proteomic analysis showed that the level of FliC protein, a component of the flagellar filaments, was lower in the culture supernatant of a spiC mutant than in the supernatant from wild-type Salmonella. Furthermore, quantitative real-time RT-PCR showed that this mutant had a much lower level of fliC mRNA, indicating that SpiC regulates the transcription of FliC. We also found that the level of SOCS-3 in J774 macrophages was lower when they were infected with the fliC mutant than with wild-type Salmonella. FliC participated in the activation of MAPK pathways in Salmonella-infected macrophages, leading to the upregulation of SOCS-3 expression. Collectively, these results indicate that SpiC is involved in the expression of FliC, which plays a significant role in the SPI-2-dependent activation of MAPK pathways in Salmonella-infected macrophages.

Involvement of Salmonella pathogenicity island 2 in the up-regulation of interleukin-10 expression in macrophages: role of protein kinase A signal pathway.

Salmonellae are facultative intracellular bacteria capable of surviving within macrophages. Salmonella pathogenicity island 2 (SPI-2) is required for growth within macrophages and for virulence in mice. In this study, we show the involvement of SPI-2 in a signal transduction pathway that induces cytokine expression in Salmonella-infected macrophages. High levels of interleukin-10 (IL-10) mRNA w...

AGE proteins as a causative factor in Alzheimer's Disease

The reaction between reducing sugars and protein free amines, known as the Maillar reaction results in the formation of advanced glycation endproducts (AGEs). AGE modification changes the structure of proteins to amyloid cross-beta structure. These protein structures can activate receptors known as RAGE on glial cells (microglia and astrocytes), and induce the expression of inducible nitric oxi...

The Jak-Stat Signaling Pathway of Interferons System: Snapshots

Interferons (IFNs) are a family of small regulatory glycoproteins that play a central role in the defense against viral infections. Although IFNs have been initially discovered as antiviral factors, today they are known as an integral part of the cytokine network that affect a wide range of biological processes. IFNs exert their pleiotropic effects through their multisubunit cell surface recept...

The Expression of Signal Regulatory Protein-alpha in Normal and Osteoarthritic Human Articular Cartilage and Its Involvement in Chondrocyte Mechano-transduction Response

Signal regulatory proteins (SIRP) belong to immunoglobulin super family (IgSF) and relate to integrin signaling cascades. It has been shown that SIRPa is expressed in a variety of cells including myeloid cells and neurons. In the present study the expression of this IgSF member in articular chondrocytes was investigated. Methods: Using a panel of anti-SIRPalpha antibodies, immunohistochemistry...
